We only put a comma after PLEASE when we make a pause in English - for instance when we implore someone very dramatically:
Please, [! pause] don't leave meeeeee!!!
Otherwise we say it with no pause: "Please call me tomorrow."

How can you tell if I made or made not a pause saying "Please, speak slower"? You cannot. But there is a pause. It is common to make a pause when starting a sentence with words like Please, Look, Listen... For example, "Look, honey, I have lost all our savings".

It may be common to make a pause in Russian, but in English, we have no pause, such as “please call me,” and the more imploring “please, call me.”

So saying/writing “please, speak more slowly” expresses frustration - as if you’ve asked this person to slow down several times and they won’t do it.

“please, speak more slowly” indeed expresses frustration and that is the main reason someone would ask to speak more slowly. 90% of cases a person would say this is because the one cannot understand or follow the speaker. Which is kind of frustration. At last, everyone can decide for oneself whether the pause exists or not. I do always have ~1 second pause when start a sentence with words like Please, Listen, Look...
